As part of a European joint project within the EraCoBiotech framework program, multi-position sensors are to be used to measure gradients in industrial scale bioreactors and mimik these gradients in laboratory scale. In particular, the influence of complex media components, especially amino acids, in interaction with gradients on bacterial and yeast cell physiology and morphology as a stress response will be investigated. In addition to investigations at the metabolome and proteome level, single cell-based optical methods are also used to evaluate vitality and viability. The results obtained will be discussed with partners from the fields of microbiology, bioprocess engineering and fluid mechanics. In addition to the experimental work, support shall be provided in conducting workshops, publishing results in scientific journals and presenting the project results at scientific conferences. In addition to work, there is the possibility of obtaining a PhD.
Test and application of mobile multiparameter probes at different reactors at industrial partners in Germany and Europe.
Replication of gradients in laboratory bioreactor cultivations and investigation of the stress response of cells at different levels taking into account the synthesis performance (biomass, proteins, secondary metabolites) using enzymatic and chromatographic detection methods, microscopic and flow cytometric investigations, partly on a single-cell level.
Evaluation of different scale-down reactors and the influence of gradients and media composition on process performance, based on this media / process and strain optimization in cooperation with the partners.
Successfully completed university degree (Master, Diplom or equivalent) in the field of biotechnology with a focus on bioengineering, the following knowledge/skills are desirable: - microbial cultivation in laboratory bioreactors, - metabolic pathways, - basic analytical methods for the determination of primary and secondary metabolites, - standard procedures in biotechnological laboratories, - presentation of scientific results.
Ideally, the candidate has experience in strain maintenance, media selection, flow cytometry and knowledge of regulatory and morphological relationships, experience in the cultivation of yeast and/or Bacillus is desirable.
The willingness to actively participate in professional events is a prerequisite. The candidate should be characterized by open communication and a good command of English (colloquial and technical language). Knowledge of German is advantageous.
